Uranium DNAzyme (ID# 7950)

n/a nM (reported value)
0.4 M NaCl and 20 mM MES buffer solution (pH 5.5) at 20?C for 15 min. To quench: 5 uL of 0.5 M Tris base solution added then sample was vortexed to shift the pH from 5.5 to 8.0.
1.0 mL 9.6 ?? 10-5 M DNAzyme 1.0 mL 6.4 ?? 10-5 M TAMRA-modified substrate Mixed with 1.0 mL of 2.0 M NaCl and 1.0 mL of 20 mM MES buffer solution (pH 5.5). Heat at 90?C for 15 min and cooled down t
Tested against Barium, Calcium, Cadmium, Chromium, Iron 2+, Iron 3+, Mercury, Magnesium, Manganese, Nickel, and Lead. Even in 100-fold excess concentrations over that used for Uranium, fluorescence intensity did not increase significantly, except in the presence of Uranium.
Limit of Detection: 0.41 nM Uranium Substrate Sequence: TAMRA-CCTC ACTAT rA GGAAG AGATG GACGT G
