On day 2, the Utrp-modified library (0.1 nmol) from round 5 of CE-SELEX was dissolved in the annealing buffer (1 mM MgCl2 in phosphate-buffered saline [PBS]). The resulting mixture was heated at 95 ◦C for 1 min, then allowed to cool slowly to 25 ◦C for 60 min to refold the aptamers.
The last four "T" bases in the secondary structure image are actually supposed to be "Utrp" as seen in the primary sequence code
