SELEX forward rounds were performed by adding 1 mg of PSMA bound to Dynabeads Talon (Invitrogen, Oslo, Norway) to 700 μl of binding buffer (100 mmol/l NaCl, 20 mmol/l Tris, 2 mmol/l MgCl2, 5 mmol/l KCl, 1 mmol/l CaCl2, 0.2% Tween-20, pH 8). The beads were removed using a Dynal magnet (Invitrogen) and were washed four times with binding buffer. At least 10 μg of ssDNA was annealed by heating to 95 °C followed by gentle cooling and was added to the PSMA matrix followed by vortexing and incubation for 1 hour at 37 °C with mild agitation.
