The DNA library was heated at 95 °C for 10 min and then gradually cooled to 25 °C over a period of 30 min in TBS buffer (10mM Tris/HCl, 150mM NaCl, 5mM KCl, 5mM MgCl2, pH 7.4).
Binding was not detectable during the course of internal investigation. Gel-Shift and Forte-Bio Octet methods to attempt to determine Kd did not generate results indicative of aptamer-target binding.
