Binding Buffer: 0.9 mM KH2P4, Na2HPO4, 70 mM NaCL, 1.3 mM KCl, 1% heparin, 0.4 g/l of streptavidin, pH 7.6.
NA If the oligo is a known aptamer sequence: For binding studies, perform a refolding protocol to ensure proper function (i.e. binding to antigen or target). Refer to the aptamer reference source for the appropriate refolding parameters and binding conditions. Note: it is unknown whether aptamer functions properly without refolding.
The aptamer did not bind to the similar human pancreatic polypeptide (hPP; ~50% sequence homology).
Truncation of the target peptide showed that this aptamer targets the C-terminus of neuropeptide Y.
Competition assays showed that this aptamer inhibits binding of neuropeptide Y to the Y2 receptor (IC50 of 73 nM). Less inhibition was observed for the Y1 and Y5 receptors (IC50 of 328 nM and 416 nM, respectively).
