The selection Buffer (SB) (0.9 mM CaCl2, 2.7 mM KCl, 1.5 mM KH2PO4, 0.6 mM MgCl2·6H2O, 0.1 M NaCl and 20 mM Na2HPO4, pH = 7.4) was purchased from Tiandz (Beijing, China). The SB was used for aptamer screening, the BLI assay, and the performance evaluation of the aptasensor.

NA If the oligo is a known aptamer sequence: For binding studies, perform a refolding protocol to ensure proper function (i.e. binding to antigen or target). Refer to the aptamer reference source for the appropriate refolding parameters and binding conditions. Note: it is unknown whether aptamer functions properly without refolding.
Specificity detection was conducted along with CTX-GI, domoic acid, okadoic acid, saxitoxin, and tetrodoxin. High response values with no significant difference were monitored when CTX-MI or the mixture of the aforesaid six toxins were added, while there was almost no response in other groups, suggesting that MBMI-01c was bound to CTX-MI with specific high affinity.
