Lectin concanavalin A (Con A-3-1m) (ID# 10082)
DNA
Lectin concanavalin A
Protein
Kd = 17nM (fluorescence particle-based assay) nM (reported value)
Incubated with Con A and FITC-PSA (counter-target, Kd = > 1 μM) in selection buffer (1xPBS, 2.5 mM MgCl2, 1mM CaCl2, 0.01% Tween 20)
Room Temperature°C
Not Reported
Counter Kd:> 1 μM (PSA). Also, ConA-3-1m showed virtually no binding to LcH, NPA, or VFA (other plant-derived mannose-binding lectins) at 10 nM
The actual aptamer crucially uses the modification 2-azidoethyl 2,3,4,6-tetra-O-acetyl-α-D-mannopyranoside at various points, which have been replaced with dTp's in the secondary structure shown (made with m-fold). This structure and the extinction coefficient may be inaccurate. The aptamer affinity was validated further through a BLI assay (Kd = 3.2 nM)
