The buffer is RPMI with 10% FBS, incubated at 37 °C (handled at room temperature). It uses 1 nmol per 10⁶ cells to detect IFNγ within 48 hours, anchored by 3′-CholTEG membrane insertion.

Resuspend at 10 µM in nuclease-free water, heat to 95 °C for 5 min, snap-cool on ice for 5 min, equilibrate at room temp for 30 min, then add to cells (1–2 nmol/10⁶ cells).
Highly specific to human IFNγ, with no response to unrelated antigens or IFNG-knockout T cells. No signal with non-target aAPCs.
