Binding assays were conducted at 37°C using PBST buffer supplemented with 5 mM MgCl₂. Aptamer concentrations ranged from 0.01 to 2.0 µM, and target VLP concentrations ranged from 1.5 to 10 µg/mL.

The aptamers were refolded by heating at 95°C for 5 minutes, followed by slow cooling to room temperature in binding buffer containing 5 mM MgCl₂ for optimal target binding.
The aptamers AP4-2 and AP17-4 exhibited high specificity for HuNoV GII.4 and GII.17 VLPs, respectively, with no significant cross-reactivity to unrelated pathogens or proteins. Specificity was validated by dot blotting and ELASA, showing strong target binding and minimal background signal (P < 0.0001).
