Molecular Activation of ST: Glycolic acid (C2H4O3, 1 g) dissolved in 4 mL of trifluoroacetic acid (C2HF3O2), 4 mL of 2.5 mg/mL ST.
Capture SELEX: 200ul of Dulbecco’s phosphate-buffered saline (DPBS), pH of 7.4
Aptamer Affinity Column: 1 mL of blocking solution (0.2% glycine solution, pH of 6.0), 1 mL of DPBS, 1 mL of PBST, and 1 mL of water.

Library and Biotin primer were mixed at a1:2 ratio and treated at 95 °C for 10 min, 60 °C for 1 min, and 25 °C for 1 min. Further, it was mixed with washed magnetic beads and incubated for 80 min at room temperature in a rotary shaker.
Based on Aptamer Affinity Column using 2.3 nmol of H Seq02, showed a binding capacity of 80 ng.
Circular dichroism and isothermal titration calorimetry further confirmed the aptamer's interaction with the target ST, supporting its potential for developing new ST detection methods.
