Protein
25 nM (reported value)
20 mmol/L HEPES (pH 7.35), 1 mmol/L MgCl2; 1 mmol/L CaCl2; and 120 mmol/L KCl

NA If the oligo is a known aptamer sequence: For binding studies, perform a refolding protocol to ensure proper function (i.e. binding to antigen or target). Refer to the aptamer reference source for the appropriate refolding parameters and binding conditions. Note: it is unknown whether aptamer functions properly without refolding.
Refolding protocol: The ssDNA pools (800 pmol for initial round) were heated at 95 °C for 5 min in a binding buffer (SHCMK) containing 20 mmol/L HEPES (N-2-hydroxyethylpiperazine-N′-2-ethanesulfonic acid), pH 7.35; 1 mmol/L MgCl2; 1 mmol/L CaCl2; and 120 mmol/L KCl; and were then cooled to room temperature. Sequence selected from library design: 5′-CGTACGGTCGACGCTAGC-N60-CACGTGGAGCTCGGATCC-3′. Full aptamer sequence should probably include library primers, but this was not specified in the reference.
