Selection buffer - 200 mM NaCl, 10 mM phosphate, 2 mM MgCl2, pH 7.6. The randomized aptamer library was annealed with the biotinylated capture strand in selection buffer. Streptavidin-agarose resin was loaded, and biotin-ssDNA complex was added into it. Bilirubin was loaded onto the library and the eluted ssDNA was obtained through gravity flow.
