RNA Nickel Small Organic 800 nM (reported value) Ni-NTA resin (200 uL) equilibrated with a buffer (50 mM NaCl, 7 mM MgCl2, 50 mM Tris-HCl (pH 7.5) was heated at 75C for 3 min and then slowly cool to RT. The Sample was loaded on the Ni-NTA matrix for 5 min under gentle agitation. N/A If the oligo is a known aptamer sequence: For binding studies, perform a refolding program to ensure proper function (i.e. binding to antigen or target). Refer to the aptamer reference source for the appropriate refolding parameters and binding conditions. Note: it is unknown whether aptamer functions properly without refolding. This may be a biphasic binding. 1-2 strong binding sites with Kd 0.8 uM, 14~16 weak binding sites with Kd 29 uM.
