Incubate 100–150 nM FAM-aptamer with 1×10⁵ target cells in DPBS (Ca²⁺/Mg²⁺, pH 7.4) + 0.05 g/L BSA + 2 µg/mL salmon sperm DNA at 4 °C for 20–30 min, then wash with DPBS.
4 °C for cell binding; 25 °C for SPR; various temps for stability tests at 37 °C
Heat the aptamer in binding buffer at 95 °C for 5 min, snap-cool on ice for 5 min, then equilibrate at 4 °C before use.
